Collagen in a cream: the molecule problem

This is not a matter of opinion or a debate between camps. The molecule is roughly six hundred times larger than the accepted limit for skin penetration, and it has been that size the entire time.

I get asked about this more than almost anything else, usually in a slightly defensive tone, because someone has already bought the cream and would like me to say something reassuring.

So let me be precise about what I am and am not saying. A collagen cream can be a genuinely good moisturiser. It can leave your skin comfortable and looking better. What it is not doing is putting collagen into the layer of your skin where collagen does its structural work. That second part is not a matter of interpretation, and it is not a fight between sceptics and enthusiasts. It is arithmetic about molecular size.

The two numbers

Collagen is a large structural protein. In its native triple-helix form, described in detail in Shoulders and Raines' review, a molecule of type I collagen has a mass in the region of 300 kilodaltons — that is 300,000 daltons.

The generally accepted working ceiling for a compound to cross intact skin is around 500 daltons. Bos and Meinardi laid out that rule in 2000, and while it is a rule of thumb rather than a law of physics — it has exceptions, and penetration enhancers shift it — the scale of the gap here does not depend on the fine print.

We are not talking about a molecule that is somewhat too large. We are talking about one that is roughly six hundred times the size of the ceiling. There is no formulation trick that closes a gap of that magnitude, and the barrier doing the excluding is functioning exactly as it should. Keeping large proteins out is not a design flaw. It is the entire job of your outer skin layer.

The second problem, which gets less attention

Suppose, for the sake of argument, that it did get through. It would still not help.

Collagen in your dermis is not a stockpile that has been deposited. It is a woven structural network, assembled in place by your own cells, cross-linked into a specific architecture. Introducing loose collagen molecules into that environment is not topping it up. The structure is made, not delivered.

This is why the same reasoning applies to elastin creams, and why I am equally unmoved by them.

So what is the cream actually doing

Something real, just not the thing on the label.

A large protein sitting on the surface of the skin behaves as a film-former and a humectant. It holds water at the surface and slows its loss. Draelos' review of moisturiser function describes this category plainly: skin that is better hydrated at the surface looks smoother and reflects light more evenly, and fine lines are genuinely less visible when the surface is plump with water.

That is a real, observable, same-day effect. It is also entirely temporary and entirely superficial, and you would get it from a well-made moisturiser containing glycerin, which costs a fraction as much.

So when someone tells me their collagen cream “definitely works”, I believe them. They are describing something they can see. They have simply been given the wrong explanation for it, and the wrong explanation is what justifies the price.

What about hydrolysed collagen and peptides

This is the part where the answer stops being a flat no, and it is worth being careful.

Hydrolysed collagen has been broken into much smaller fragments. Some of those are small enough that penetration is at least plausible rather than physically excluded. And there is a class of short chains — signal peptides, a few amino acids long — with a legitimate research literature behind them, which appear to act as messengers that may encourage the skin's own cells to produce more collagen.

Note what changed. That is not collagen going in. It is a small fragment that may prompt your own machinery to build more. Entirely different mechanism, entirely different claim, and one I take seriously enough that it deserves its own discussion rather than a paragraph here.

It is also why an ingredient list matters more than a product name. “Collagen” on the front tells you almost nothing about which of these three quite different things is in the bottle.

Ingesting it is a separate question

Collagen drinks and powders are a different conversation with a different and more mixed literature, and I am not going to fold them into this one. Digestion breaks the protein down into amino acids and small peptides, which is a completely different pathway from smearing it on. Whatever you conclude about the drinks, it has no bearing on the cream.

What I would do

If the cream feels good and you like it, keep using it — as a moisturiser, at a moisturiser's price, with a moisturiser's expectations. Judge it by whether your skin is comfortable, not by whether it is rebuilding anything.

If you are buying it specifically for firmness, you are paying a premium for a mechanism that is not occurring, and I would rather you spent that money on the boring things with better evidence behind them. Sunscreen, used in the right amount, has more support for preserving the collagen you already have than any cream has for adding to it.

That is the unglamorous version. It is also the one that holds up.
